摘要: |
絡(luò)石藤為藥典記載的常用中藥,而地瓜藤在民間部分地區(qū)用作絡(luò)石藤。為明確兩種藥材抗炎活性及相關(guān)化學(xué)成分間的差異,初步探討其可能的藥效物質(zhì)基礎(chǔ),該研究通過(guò)檢測(cè)不同濃度絡(luò)石藤與地瓜藤醇提物對(duì)RAW264.7細(xì)胞存活率的影響,采用Griess試劑法測(cè)定LPS誘導(dǎo)的RAW264.7細(xì)胞上清液NO釋放量,利用UPLC-Q-TOF-MS/MS技術(shù)對(duì)絡(luò)石藤與地瓜藤醇提物中的化學(xué)成分進(jìn)行分析,并結(jié)合多元統(tǒng)計(jì)學(xué)中的OPLS-DA模型鑒定兩種藥材的差異性化學(xué)成分。結(jié)果表明:(1)兩種藥材皆具有一定的NO抑制作用,絡(luò)石藤粗提物的抑制作用稍強(qiáng)于地瓜藤。(2)絡(luò)石藤與地瓜藤的化學(xué)成分存在明顯差異,絡(luò)石藤中的21個(gè)差異性成分主要為木脂素類(lèi),而地瓜藤中的10個(gè)差異性成分主要為黃酮類(lèi)。綜上認(rèn)為,差異性化學(xué)成分可能是造成這兩種藥材抗炎活性差異的主要活性物質(zhì)。該研究結(jié)果為絡(luò)石藤與地瓜藤的抗炎藥效物質(zhì)基礎(chǔ)和質(zhì)量控制提供了理論依據(jù)。 |

Inhibition of NO activity and differential chemical constituents of the medicinal parts in Trachelospermum jasminoides and Ficus tikoua |

Abstract: |
Trachelospermum jaminoides, a liana of family Apocynaceae, is used as classic traditional Chinese medicine recorded in the Chinese Pharmacopoeia, and Ficus tikoua, a liana of family Moraceae, is used as its customary supply in some ethnic districts, both of which are used as remedies for the inflammation-related disease. In order to determine the differences of anti-inflammatory activity and related chemical constituents between the two medicinal materials, the cell viability test was used to detect the cytotoxicity(RAW264.7)of the medicinal parts in T. jaminoides and F. tikoua at different concentrations, and the Griess method was applied to detect the inhibitory effects of the two species on nitric oxide(NO)production in LPS-induced RAW264.7 cells in vitro. The ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS)combined with the OPLS-DA model in multivariate statistics were used to identify the differential chemical constituents between the medicinab parts in T. jaminoides and F. tikoua. The results were as follows:(1)The NO production inhibition rate of T. jaminoides is stronger than that of F. tikoua.(2)Twenty-one differential chemical components were identified in the stems with leaves of T. jaminoides, most of which were lignans. Ten differential chemical components in the aerial parts of F. tikoua, most flavonoids, were identified. In summary, the differential chemical constituents may be the main active substances that cause the difference in anti-inflammatory activity between the two medicinal materials. This study provides a theoretical basis for the anti-inflammatory medicinal material basis and quality control of the medicinal parts in T. jaminoides and F. tikoua. |
